A comprehensive inventory for phenotype characterization - Interview for Psychiatric Genetic Studies. This included the Structured Clinical Interview of DSM IV Axis I Disorders Interview for Diagnostic and Statistical Manual Of Mental Disorder-IV and the OPerational CRITeria system as well as review of medical records. |

Statistical Method |
Departure from Hardy-Weinberg-equilibrium was tested using a X2- test (1df). Single-marker association analysis was performed using the Cochrane-Armitage Trend Test. Power calculations were conducted with the Genetic Power Calculator. The haplotype distributions of cases and controls were compared using FAMHAP. All marker combinations were tested according to Becker et al. (2005a) and a global P value to correct for multiple testing was computed using Monte-Carlo simulations. |
Result Summary |
Although no association was found for VAMP3 markers, we observed evidence of association with SNPs at the VAMP2 locus in the initial sample (P values between 0.005 and 0.033). To validate these findings, we analyzed a second BPAD sample and failed to replicate the initial findings at the single-marker and haplotypic level. Our results do not suggest that a common genetic variant at VAMP2 or VAMP3 contributes to the development of bipolar affective disorder in German patients. |

For initial sample, corrected global P-value = 0.013, the strongest association was obtained for the haplotype G-A at markers rs2278637-rs8067606, which occurred with a frequency of 35.8% in patients and 29.2% in controls. For replication sample, global P-value = 0.018, haplotype G-G (at rs2278637-rs8067606), which showed opposite tendencies in the initial sample. The former observed risk-haploypte G-A at the same marker combination occurred more frequently in controls (34.0%) compared with the patients (31.9%) in the replication sample.
