Sample Status	Subjects were recruited through the COSMO (Collaborative Study of Mood Disorders) consortium, which comprised of 867 unrelated bipolar disorder patients (588 bipolar disorder I, 279 bipolar disorder II) and 895 age- and sex-matched controls. The patients were diagnosed by at least two experienced psychiatrists according to the DSM-IV criteria for bipolar disorder I or bipolar disorder II on the basis of unstructured interview and reviews of their medical records.
Technique	genotyping was performed using TaqMan assay; PCR was performed using an ABI 9700 thermocycler, and fluorescent signals were analyzed on an ABI 7900HT Fast Real-Time PCR System using Sequence Detection Software (SDS) v2.4 (Applied Biosystems).
Statistical Method	Genotype and allele frequencies were computed and were checked for deviation from Hardy-Weinberg equilibrium. Fisher's exact test (two-tailed) was used to compare allele frequencies between patients and control subjects. Statistical differences in genotype distributions were evaluated using Pearson's _2 test.
Result Summary	Results showed no significant differences in allele and genotype frequencies of EGR2 SNPs between bipolar disorder patients and controls and also in a sex-stratified genetic analysis. The haplotype and meta-analyses also showed no significant association with bipolar disorder. In conclusion, this is the first replication study of the previously reported association of EGR2 with bipolar disorder in a larger sample set and the results showed that the EGR2 gene is unlikely to contribute to the susceptibility of bipolar disorder in a Japanese cohort.
