| Disease Type | Bipolar I Disorder |
| Study Design | family-based |
| Study Type | Candidate-gene association study |
| Sample Size | 170 bipolar-I affected offspring triads were drawn from 522 individuals with 2 parents in 332 nuclear families recruited for genetic studies. |
| SNP/Region/Marker Size | 28 SNPs |
| Predominant Ethnicity | Caucasian |
| Population | European |
| Gender | 124 males, 94 females for bipolar offspring |
| Age Group | Children : Bipolar offspring mean age=12.9 (SD=5.3) years old, |
| Sample Diagnosis | DSM-IV |
|---|---|
| Sample Status | All children met criteria for DSM-IV bipolar I disorder. The mean age at onset was 7.3, SD=4.8 years of age and the clinical presentation was chronic (3.5 SD=3.6 years duration), rapid cycling (18.4 SD=53.5 episodes), and functionally impairing (GAF score of 41.0 SD=6.2). In addition, 85% (N=145) of children with bipolar-I disorder also met criteria for ADHD. In addition, 45 affected:unaffected parent pairs were identified. Parents with bipolar-I disorder had an average age at onset of 23.1 SD=10.9 years and duration of 5.9 SD=7.8 years of illness. |
| Statistical Method | For the Transmission Disequilibrium Test (TDT) we used the PLINK (v1.0) software package. Odds ratios and 95% confidence intervals, however are calculated using only the basic TDT. Multiple permutation (n=100,000) was used to calculate empirical P values and to correct for the number of statistical comparisons conducted. Findings were considered statistically significant at permuted P<0.05. |
| Result Summary | Results indicated nominally positive association for 4 SNPs (rs40184, rs11133767, rs3776512, and rs464049), but only rs40184 survived correction formultiple statistical comparisons (P=0.038). These data provide suggestive evidence supporting a role for SLC6A3 in the etiology of pediatric bipolar disorder. |
