| Sample Size |
The subjects investigated were from 312 nuclear families, each with at least one proband (118 males and 194 females) affected with DSM-IV BD type I or BD type II. In addition, 26 siblings with BD were included in the family-based analysis. The total sample consisted of 1043 subjects: 350 BD patients and 693 unaffected relatives. |

The total sample consisted of 350 BD patients (131males and 219 females). |

| Technique |
qPCR; BDNF Val66Met genotyping was performed using commercially available TaqMan? allelic discrimination assays on the ABI 7000 Sequence Detection System (ABI), following the manufacturer's protocol. |
| Statistical Method |
Association tests and transmission disequilibrium test(TDT) analyses of the BDNF Val66Met SNP and BD were performed using TDTPHASE. Differences in effect size between maternal and paternal transmission ratios were assessed by calculating Pearson's X2 values. |
| Result Summary |
The BDNF Val66 allele was transmitted significantly more often to patients with BD (maternal transmissions: 46/22, p = 0.003; paternal transmissions: 55/30, p = 0.006). There was no significant difference between maternal and paternal transmission ratios. There was no significant difference in the ratio of Val/Met-specific mRNA expression between BD and controls, in either brain or B lymphoblasts. The Val/Met ratiowas much lower in the brain vs. B lymphoblasts. |
