| Citation | Roche, S. and P. McKeon (2009). Support for tryptophan hydroxylase-2 as a susceptibility gene for bipolar affective disorder. Psychiatr Genet 19(3): 142-146. |
| Disease Type | Bipolar Disorder |
| Study Design | family-based |
| Study Type | Candidate-gene association study |
| Sample Size | 151 BPAD type I trios |
| SNP/Region/Marker Size | 13 SNPs |
| Predominant Ethnicity | Caucasian |
| Population | Irish |
| Sample Diagnosis | DSM and SCID |
|---|---|
| Sample Status | All trios are of Irish descent and were diagnosed based on the Diagnostic and Statistical Manual of Mental Disorders, fourth edition (DSM-IV) criteria and using information gained from hospital records and semistructured interviews conducted using the Schedule of Affective Disorders and Schizophrenia (Lifetime version; SADS-LB) or Structured Clinical Interview for DSM-IV Axis I Disorders: Clinical Version (SCID-CV) interview schedules. |
| Technique | genotyping |
| Statistical Method | Transmission Disequilibrium Test (TDT) |
| Result Summary | SNPs rs1386482 and rs1386486, which are in very strong linkage disequilibrium, were associated with BPAD (P=0.006). The association retained significance after a correction for multiple testing. The associated SNPs are in perfect linkage disequilibrium with SNPs previously associated with BPAD (rs4290270) and impulsivity (rs1386483), a core trait of BPAD. These results strongly support a role for TPH2 in the aetiology of BPAD. |
